Chardy, ranked 58 in the world, beat Germany’s Florian Mayer 6-2 7-6(7-4) in Tuesday’s first round encounter.
Murray’s won his only previous match against Chardy in Montreal last year in straight sets.
The Scot comes into this week’s tournament on a high after his win over Roger Federer in last weekend’s Rogers Cup final in Toronto.
He is aiming for success at the US Open, the year’s final Grand Slam, which begins on 30 August in New York.
But there were defeats for the 12th seed Mikhail Youzhny and 16th seed Gael Monfils.
Youzhny fell to Frenchman Richard Gasquet 2-6 6-4 6-1, just after Monfils was upset by Alejandro Falla of Colombia 6-3 6-4.
Former world number four James Blake also lost out with his conqueror Denis Istomin of Uzbekistan to face Roger Federer in the next round but Blake’s compatriot Mardy Fish beat Gilles Simon 7-6 7-5 and set up a match with Spanish eighth seed Fernando Verdasco.
Lleyton Hewitt advanced after Taiwan’s Lu Yen-Hsun retired when 6-4 4-0 down while Wimbledon runner-up and seventh seed Tomas Berdych of the Czech Republic beat Paul-Henri Mathieu of France 6-4 6-4.
